Advances in single-molecule spectroscopy and biological applications

Single-molecule spectroscopy is a unique tool which can provide both high spatial resolution and high temporal resolution under biologically relevant conditions and has been invaluable in determining the detailed kinetics and mechanisms of biological processes. In order to make these approaches more powerful and broadly applicable, we’ve pioneered three-color single-molecule FRET (fluorescence resonance energy transfer), combination of single-molecule FRET and mechanical manipulation, and suppression of quantum dot blinking. I’ll also discuss some of the many possible biological applications of these new technical advances and future research directions.
